| Abstract: | This study aimed to evaluate magnetic hyperthermia (MHT) mediated by magnetic nanoparticles as a potential method to induce infertility in male cats. A total of 20 cats were divided into two experimental phases to assess short-term effects – Phase 1 (N = 11) and long-term effects – Phase 2 (N = 9). The treatment consisted of intratesticular injection of 150 µL of a magnetic fluid composed of citrate-coated manganese ferrite, followed by MHT until testicular temperature reached 45°C, maintained for 15 minutes. Orchiectomy was performed at different time points post-MHT – Phase 1: (Group D7, N = 3; Group D24, N = 4; Group D47, N = 4) and Phase 2: (Group D90, N = 2; Group D145, N = 3; Group D180, N = 4). Clinical, hematological, and ultrasonographic parameters were evaluated at various pre- and post-treatment intervals to monitor animal health and welfare. MHT was found to be safe and well-tolerated, with no significant increase in testicular temperature, no marked clinical signs of pain, and no systemic compromise. Among the testes evaluated post-treatment, 57.5% were firmer within the first 24 hours, 65% showed increased sensitivity within the first 2 hours, and 85% presented scrotal erythema within the first 4 hours. These alterations were no longer observed from day 7 onward. A statistically significant reduction in absolute testicular weight was observed on day 180 compared to the control group. Testicular volume increased significantly on day 7 due to peritesticular inflammation, followed by a marked decrease on day 180. Azoospermia was observed in 80% of the treated animals. Histological analysis revealed substantial damage to the architecture of seminiferous tubules and epididymides in both phases, depending on the intensity of the inflammatory process and the presence, quantity, and proximity of nanoparticle-like material. The presence of nanoparticles was associated with cellular damage, particularly in areas of higher accumulation, leading to diffuse necrosis of testicular tissue. These findings indicate the potential of the proposed method as a non-surgical alternative for sterilization in male cats. |
