| Abstract: | Urinary tract infections (UTIs) are among the most common infections in clinical practice and represent a major public health concern. Estimates indicate that, in 2019, approximately 400 million cases of UTIs were reported worldwide. Frequently caused by bacteria such as Escherichia coli, Klebsiella spp., Proteus mirabilis, and Staphylococcus saprophyticus, these infections have become increasingly associated with the rise in antimicrobial resistance. The time required for diagnosis using conventional methods is considerable, often leading to the empirical initiation of treatment, generally with broad-spectrum antibiotics; this practice contributes to the increase in antimicrobial resistance, which currently represents one of the main public health problems worldwide. Considering the need for faster diagnostic methods, this study investigated the ability of three polyclonal antibodies, developed from three distinct epitopes of S. saprophyticus cell wall-associated proteins, to recognize other uropathogenic bacteria. To this end, isolates of E. coli, K. pneumoniae, K. variicola, and P. mirabilis were subjected to an adapted On-Cell Western protocol, which involves incubation with primary and secondary antibodies directly on intact cells, without the need for protein extraction, followed by chemiluminescent detection. The results demonstrated that the antibodies were able to recognize E. coli, K. pneumoniae, and K. variicola, suggesting that the epitopes used are structurally conserved among these species. The experimental evaluation of P. mirabilis, in turn, required cautious interpretation due to the presence of signal in the negative control. When applied to Candida albicans, a microorganism that may be part of the human microbiota and act as an opportunistic uropathogen under conditions of immunological imbalance, no recognition was observed, reinforcing the specificity of the antibodies for antigens of bacterial origin. Taken together, these findings highlight the relevance of an antibodybased immunodiagnostic approach as a promising strategy for the detection of uropathogenic bacteria and for the future development of faster diagnostic methods for UTIs, with potential contributions to improved clinical management and to the promotion of the rational use of antimicrobials. |
