| Abstract: | Cryptococcosis is a systemic mycosis caused by yeasts of the genus Cryptococcus, and is one of the main invasive mycosis in humans. In recent years, an increase in the incidence of fungal infections has been observed, especially in patients with some type of immunocompromise. Several studies have shown that the increase in capsule size or formation of titan cells of Cryptococcus spp. may be involved in crucial processes of the initial adaptation of the fungus to vertebrate and invertebrate hosts. Both processes reduce the recognition and/or phagocytosis of the fungus and other mechanisms involved in the destruction of the fungus by the immune system. In addition, the formation of titan cells, which are capable of producing daughter cells with the ability to change their size, are possibly involved in the latency processes of this opportunistic pathogen. Titan cells are a potential target for studies of parasite-host interactions and are already cited as an important virulence factor in cryptococcosis. Our group had demonstrated that incubation of C. neoformans cells with phospholipids induces not only an increase in the size of the capsule of this fungus, but also the formation of titan cells. Thus, the objective of this study was to analyze how physical and chemical conditions can affect how phospholipids induce capsula enlargement and titan cell formation in C. neoformans. We observed a greater induction of fungal capsule size after incubation with the phospholipid L-α-phosphatidylcholine with a higher degree of purity, and when the assay was conducted in 96-well plates for 72 hours at 30°C, under agitation. Regarding the formation of titan cells, titan cells were obtained in all evaluated conditions when we added the phospholipid P1; however, using the phospholipid with a lower degree of purity, in some cases, we did not observe a significant increase in cell body size. We did not observe effects related to growth at temperatures of 37°C or 30°C, nor in the presence or absence of a film for hypoxia induction in the cultures. These results help clarify the role of phospholipids in inducing capsule enlargement as well as titan cell formation in C. neoformans. However, we observed that other factors need to be studied further for a better understanding of elements important in inducing these phenotypes in C. neoformans in response to phospholipids. |
