Abstract: Lettuce is a vegetable widely distributed throughout the country, representing one of the most cultivated plants and the most consumed leaf in Brazil and in the world. However, the lettuce production system in Brazil is affected by several environmental factors, diseases and pests that cause great damage to producers. In this context, the genetic transformation of plants stands out as an important biotechnological tool and has been employed for the most diverse purposes. However, success in the plant transformation process is associated with the use of stable promoters. Being reported in lettuce a difficulty in maintaining the stability of transgene expression, possibly due to the use of the 35SCaMV promoter. In the first chapter the objective of this work was to test two new promoters, aiming at alternatives for lettuce genetic transformation works. Twelve, 8 and 5 genetically modified strains expressing the gus gene were obtained under the control of the pACT2, pAVP1 and p35SCaMV promoters, respectively. Each plant was analyzed by PCR and histochemical assay in each generation (T0, T1 and T2). GUS activity was detected in the leaf tissue of all transgenic (T0) plants obtained with pACT2 in 3 of 8 pAVP1 plants and 2 of 5 p35SCaMV plants. The transgenic plants were also submitted to fluorimetric assay to quantify GUS activity. In the second chapter the objective was biofortification for folate increase in genetically modified lettuce plants. Agrobacterium tumefaciens mediated lettuce genetic transformation experiments were carried out to obtain genetically modified strains expressing the gchI and adcs genes, under the control of the ACT2 and AVP1 promoters, respectively. Three transgenic strains were confirmed after PCR analysis. The transgenic strains obtained showed an increase of up to 6.3 times more total folates when compared to non transgenic lettuce (control). And up to 2.5 times more folate compared to spinach (Spinacea oleracea), which is a folate-rich plant.
