| Abstract: | Hydrogels formulated from polysaccharides have gained great importance in many fields of application, being used from personal hygiene products to controlled drug release devices and tissue repair in human body. Classified as biocompatible, hydrogels obtained from natural sources are able to absorb large amounts of water and biological fluids, thus being their main application in medical and pharmaceutical materials. However, they need to take careful steps to eliminate toxic components, whether in obtaining the raw material or the final product. This work aims to optimize the purification of a natural hydrogel obtained from Magonia pubescens - typical fruit from Brazilian Cerrado - through two distinct methods: dry and wet separation of hydrogel from biomass; followed by oxidative purification and chemical modification with N'-N-methylenebisacrylamide (MBA), so that the product could absorb water more than 150 times its own mass. Dry Purified Modified (BMES) and Wet Modified (BMEU) Biogels were evaluated by FT-IR, TG / DTG / DSC, SEM and swelling degree. The purification study method employed was better suited to the suggested applications when using hydrogen peroxide (H2O2) solution at pH 10, since they produce less toxic or undesirable waste and byproducts, and do not change the absorption characteristics of water from biogel, turning it into a homogeneous and translucent product. FT-IR analyzes revealed the incorporation of MBA into the polymeric matrix showed the appearance of characteristic amide group bands and TG / DTG / DSC resulted in typical thermograms of cellulosic materials. The textural analyzes also showed the fibrous and porous appearance of the material, as well as the emergence of new structures attributed to the self-healing of the crosslinking agent. However, to water absorption capacity, chemical modification has not yet been found to be suitable for BMEU, causing a loss in absorbency. In feasibility study of use these materials in promotion of human health, unmodified Biogel was evaluated for its toxicity and as a drug release device, being classified as non-toxic according to standards established by the World Health Organization (WHO). As for drug release, it allowed 90% of diclofenac sodium and 70% of Amphotericin-B to be released from its structure, becoming available for skin absorption, thus being a potential substitute for application as a vehicle for incorporation of topical drugs. Digestibility simulations were also applied to the materials, demonstrating that they can be studied as auxiliary devices for weight control and reduction because, in stomach solution, they have swelling and little dissolution, promoting satiety to the patient for a long time. When designed for use on a cosmetic or pharmacological basis, biogels were subjected to shelf tests suggested by ANVISA to determine their average durability without losing their initial physicochemical characteristics. For this, organoleptic characteristics such as color, appearance and odor were evaluated as well as pH and density in controlled environments. In the tests, both gels (BEU and BES) presented durability of less than 90 days, presenting biological contamination attested by the emergence of fungi and odor alteration. Thinking about the presentation of this product, the formation of films with wet and purified biogel (BEU) was tested and, after adaptations with glycerol as plasticizer, the result was a high quality film, arousing interest in its optimization for several applications. |
