Abstract: Monoconidial cultures of 46 strains of Trichoderma isolated from brazilian cerrado soils were characterized on the basis of morphological and cultural characteristics, and on RFLP of the internal transcribed spacer (ITS 1 and 2, including 5.8S gene). The most common group comprised 34 isolates and was identified as T. harzianum. The remainder isolates clustered into three groups: T. asperellum (4 isolates), T. koningii (3 isolates), and T. spirale ? (2 isolates). In addition, three isolates with smooth ellipsoidal conidia, and pronunced coconut odour may be an undescribed taxa; although the digestion analysis of the rDNA these isolates with Eco RI, Hha I, Bam HI, and Msp I have showed the same pattern of the T. koningii group. Morphological variation was noted specially among isolates of T. koningii, and more subtle variation were also observed for T. harzianum. Genetic diversity using RAPD-PCR, biological activity against R. solani, and biochemical features were also assessed for T. harzianum isolates. Considerable genetic variation was noted among isolates of T. harzianum. Most of the T. harzianum tested are able to produce chitinases and p-1,3-glucanases when grown in chitin containing medium and presented different levels of mycoparasitic activity against R. solani. Similarities between levels and specificities of biological activity and the DNA-analysis-based groupings are both discussed in relation to T. harzianums potential as a biocontrol agent.
