Abstract: Introduction: The classification of insecticide-resistant sandfly populations is based on concepts and methodologies used to characterize the susceptibility profile in mosquitoes. However, sandflies are insects with distinct biology which could lead to erroneous and subjective interpretations in classifying a population to insecticides. Objective: To evaluate the impact of biological variables on the susceptibility of the sandfly Lutzomyia longipalpis to the alpha- pipermethrin insecticide, as well as to identify and describe the reports of sandfly resistance to insecticides in the world and suggest a reference population for future biological tests. Methodology: Lu. longipalpis species were evaluated in relation to sex, age, nutritional status, mortality criteria and sub-lethal effect in the methodologies of CDC bottles and modified WHO cones for the insecticides alpha-cypermethrin and bendiocarb and the diluents alcohol and acetone. In addition, 61 studies, obtained systematically from four scientific databases, were included for evaluation. In addition, three sandfly populations were analyzed for the designation of a reference population. Results: The Kaplan-Meier survival curves showed a statistical difference in the diet of sandflies, in the bottle methodology, where females fed with blood were more susceptible when compared to females fed only with sugary solution using the bottle method (p = 0.001) and age, in the modified cone methodology, in which five-day-old sandflies were more susceptible than three-day-old sandflies (p = 0.001). Gender, alcohol and acetone did not influence the results of biological tests. Records of phlebotomine resistance are described for all classes of insecticides, mainly for the species Phlebotomus papatasi and Ph. argentipes, from the Old World. However, only two studies carried out comparisons between field phlebotomines and xx phlebotomines kept in the laboratory (control) and the phlebotomine doses and lethal times are very varied, revealing that there is no specific protocol for assessing the phlebotomine susceptibility to insecticides. Conclusions: It is concluded that the susceptibility of sandflies to insecticide is influenced by age and nutritional status. However, the sex of insects does not seem to impact the analysis. Currently, there are 47 populations of Phlebotominae with confirmed resistance to one or more insecticides and another 28 populations considered tolerant. In addition, the population of Lu. longipalpis originating from Jacobina and created at Fiocruz in Rio de Janeiro, here it is suggested as a Brazilian Susceptibility Reference Lineage.
