Abstract: Spiders are arthropods from the Aranae order that have in their venoms a great diversity of compounds, such as salts, organic acids, enzymes, proteins and peptides. Among these compounds, peptides are of pharmacological interest due to their neurotoxic activity, acting on voltage-dependent ion channels present in the central nervous system, peripheral nervous system, cardiac muscle and skeletal muscle. In the venom of the spider Acanthoscurria paulensis found in Brazilian territory, approximately 100 components were described, among these components only the peptide toxin Ap1a was purified and had experiments to characterize biological activity. In this present work, five new peptide toxins entitled as Ap2, Ap3, Ap4, Ap5 and Ap6 were purified by RP-HPLC, sequenced by In-souce Decay and their sequences completed and confirmed by analysis in the transcriptome of the venom gland. A. paulensis. The Ap2, Ap3, Ap4, Ap5 and Ap6 peptides have, respectively, 42, 41, 35, 46 and 31 amino acid residues, and experimental molecular masses of 4886.3, 4883.7, 3870.76, 5454.9 and 3717.9 Da, with the Ap2 peptide having the amidated C-terminal serine. Searching in sequence databases indicated that Ap2, Ap3, Ap4, Ap5 and Ap6 peptides have similarities with peptide toxins that present the structural motif known as ICK (Inhibitory Cystine Knot), which have the possibility of modulating ion channels. The toxins isolated in the present work were submitted to electrophysiological experiments in voltage-dependent Na+ (NaV) and Ca2+ (CaV) channels. Among the channels NaV1.1, NaV1.5, NaV1.7, CaV1.2, CaV2.1 and CaV2.2 tested, the toxins Ap2, Ap3, Ap5 and Ap6, showed inhibition activity between 10 and 30%, in CaV2.1 channel. These four toxins are, therefore, characterized as the first ω-toxins of spiders of the genus Acanthoscurria that have activity in calcium channels described. The Ap3 toxin inhibited the channels NaV1.1, NaV1.5 and NaV1.7 by 6.6%, 4.2% and 12.05% respectively. With these results, the present work contributes to the expansion of knowledge about the components of the venom of spiders in Brazil, as well as to the description of new bioactive compounds that can be used as pharmacological tools to understand the mechanism of functioning of ion channels and interaction with peptide toxins.
