Abstract: Chromatin is the major regulator of gene expression and genome maintenance. Its structure consists of nucleosomes, formed by histone octamer (H2A, H2B, H3 and H4) wrapped by DNA of 145-147bp. The regulation of chromatin structure is an extremely dynamic phenomenon and involves several factors and mechanisms. In addition to the nucleosome binding proteins, which are determinant to dictate the chromatin outcome, it is plausible to consider that small molecules and peptides present in the nuclear compartment have important action on the nucleosome structure and, consequently, on the chromatin dynamics. In this work we aimed to study: i) impact of cholesterol on chromatin structure and ii) effect of nucleosome ligand peptides on chromatin in vitro and in cell based assays. In the first part of this work, we observed that the binding of cholesterol to chromatin assists the formation mononucleosomes and 30nm fibers in vitro, as well as increases stability and induces compaction of long chromatin fibers. Our approach suggests that a lipid, such as cholesterol, can be physiologically considered as a nucleosome ligand molecule with influence on chromatin structure. In the second part, we demonstrate that the peptide designed in silico binds to the nucleosomes in vitro and does not affect its stability. We observed that the peptide induces long chromatin fibers compaction in the presence of magnesium in vitro. Analyzes by confocal laser microscopy revealed that the peptide is able to penetrate in Hela cell nuclei in five minutes. Cytotoxicity assays revealed that the peptide decreased the viability of tumor cells (Hela, Raw 264.7 and C3H10 T1/2), which did not occur with the normal cell line (CCD1059). In addition, analyzes of the relative expression of the TNFα gene demonstrated that the peptide caused up to five-fold reduction in the expression of that gene. Thus, we believe that modulation of chromatin by exogenous nucleosome ligand molecules may be a valuable mechanism from a pharmacological point of view, and can be used to regulate various cellular disorders.
