Abstract: Infections caused by human herpesviruses are of an important public health concern with a high world population prevalence. Therapy for such infections is carried out by nucleoside analogues, which target viral DNA polymerase. However, the prolonged drugs utilization can trigger the resistant variants emergence, leading to severe infections. Therefore, it is imperative the study of molecules capable of acting on different viral or cellular targets with low host toxicity. This study aimed to evaluate the in vitro action of molecules derived from natural products and immunomodulatory peptides against HHV-1 replication, as well as to infer their possible effect on stages of the viral replicative cycle. The cytotoxic effects of such molecules onto Vero cells were evaluated by MTT method. The chalcone cardamonin showed toxicity at concentrations above 25 μg.mL-1. Moreover, for the peptide DJK-6, there was cell viability reduction at concentration of 200 μg.mL-1 and IDR-1018 did not present cytotoxicity. In the antiviral assays, cardamonin showed complete CPE viral inhibition in MNTC (25 μg.mL-1) with SI around 21.8. In addition, activities over penetration and intracellular assay were observed. IDR-1018 and DJK-6 showed viral inhibition above 90 % in MNTCs (200 μg.mL-1 and 100 μg.mL-1) and SI of >22.9 and >8.2, respectively. Regarding the effects on the viral replicative cycle, IDR-1018 showed activity in adsorption, penetration and intracellular assays. Otherwise, DJK-6 showed activity only over penetration and intracellular viral stages. This study indicated that such molecules tested may be promising for anti HHV-1 new drugs development due to their inhibitory activity in vitro and the ability to act at different viral replicative cycle stages. The chalcone cardamonin showed inhibitory activity against viral replication at small concentrations. In relation to the immunomodulatory peptides, the IDR-1018 analog present higher potency and lower cytotoxicity compared to DJK-6, in addition to a better safety profile.
