Abstract: The tomato (Solanum lycopersicum L.) is one of the main vegetables grown in the world. The cultivation of tomato represents an important economic and social activity in Brazil. Viruses represent a major phytosanitary problem for the major growing regions of this vegetable, being the begomovirus, tospovirus and crinivirus the ones with great importance and high incidence. Recently, there were reports of the occurrence of a tobravirus in Brazil and the lack of information about this virus disease justifies a high demand on starting a research on this important virus group. Members of the genus Tobravirus have the genome constituted by positive sense RNA in a bipartite genome (RNA1 and RNA2). Tobraviruses are transmitted plant-by-plant by soil nematodes of the genus Paratrichodorus and Trichodorus (family Trichodoridae), considered as their natural vectors. This genus is represented by only three species, Tobacco rattle virus (TRV), Pea early-browning virus (PEBV) and Pepper ringspot virus (PepRSV). Known as ‘vírus da mancha anelar do pimentão’ or ‘vírus da faixa amarela’, PepRSV is only described in Brazil, causing diseases in pepper, artichoke, patchuli and tomato plants. The first isolate characterized was from pepper and is known as isolate CAM (Campinas-SP). In 2012, the occurrence two isolates of PepRSV was reported in processing tomatoes in Luziânia, GO, denominated LAV and Pivo4 . The general objective of this work was to study the interaction of PepRSV with the host. Two isolates were used, CAM from pepper and LAV, collected in tomato in 2012. Initially, these isolates were characterized biologically and an antibody was produced (Chapter 2). Furthermore, infectious clones were constructed (Chapter 3). In the chapter 2, the host range was analyzed afer mechanical inoculation in indicator plants, and tomato cultivars. Viral particles were purified for the production of antibodies in rabbits. The presence of the purified particles was confirmed by transmission electron microscopy, and an atibody with good specificity was produced. In the chapter 3, PepRSV infectious clones were constructed by extracting total RNA, followed by an RNA-1 and RNA2 RT-PCR that were inserted into the pJL-89 cloning vector by Gibson Assembly and later were agro-infiltrated into susceptible plants. It was concluded that it was possible to produce anti-CP polyclonal antibody; the PepRSV CAM isolate infects 16 different plants systemically; PepRSV isolate CAM translocates to the root system when inoculated on the aerial part of the hosts Nicotiana tabacum 'Samsun', N. tabacum 'TNN', N. rustica, Capsicum chinense PI159236, C. annuum 'Ikeda', S. lycopersicum cv. Santa Clara, Chenopodium quinoa and Gomphrena globosa; the tomato cultivars AP533, N901, U2006, Money Maker and Gaucho were susceptible to infection by PepRSV isolated CAM; a new PepRSV isolate named Ag1 was detected; and finally the clones of PepRSV were infectious to N. benthamiana plants.
