Abstract: Whilst carbohydrates are the principal respiratory substrate in plants, under certain circumstances amino acids may be catabolised in order to produce energy, reducing equivalents and precursors for biosynthesis. This situation may occur during the phase of reserve mobilisation in seedlings, and this period of development therefore represents an opportunity to identify which amino acids are in fact degraded. In this study, we therefore carried out an analysis of amino acid balance in Vicia faba L. seedlings by quantifying both free and protein bound amino acids at two time periods. Comparison of the total amounts of amino acids points to the catabolism of at least eight amino acids, whilst the amounts of others remained constant during the period analysed. GC–MS-based metabolite profiling detected several intermediates of catabolism and suggested that carbon may be directed from amino acid catabolism into the TCA cycle. The pathways by which two of the amino acids, histidine and phenylalanine, are catabolised have received limited study, suggesting that this may represent a useful experimental system for further investigation of these processes.
