| dc.description.abstract | O uso de cogumelos como agentes benéficos à saúde humana remonta a milhares de
anos, especialmente na cultura asiática. A prospecção do uso de polissacarídeos, investigados
como potenciais mediadores de diferentes respostas imunológicas associadas a sistemas de
inflamação e infecção tem sido foco de pesquisa nos últimos anos. Estudos recentes
demonstram que o basidiomiceto Auricularia auricula, considerado o terceiro cogumelo
comestível de maior importância cultivado no mundo, é utilizado não só no contexto
alimentício, como alimentos funcionais, mas também apresenta valor para a indústria
farmacêutica por seus compostos bioativos, como os polissacarídeos, vitaminas, proteínas,
polifenóis e minerais. Polissacarídeos obtidos deste cogumelo apresentam atividade
antioxidante, antitumoral, anticoagulante, hipoglicêmica, radioprotetora, antiviral e
imunomodulatória.
A modulação da resposta imune por polissacarídeos é bem descrita na literatura, com a
atuação de receptores celulares responsáveis pela ligação de açúcares que levam a ativação e
sinalização celular, desencadeando ativando resposta imune inata e adaptativa. A característica
da resposta, entretanto, está associada com a composição molecular e estrutural destas
moléculas, podendo variar com a composição monossacarídica, padrão de ramificação, peso
molecular, solubilidade, método de extração da molécula e o grau de pureza obtido.
Um dos polissacarídeos caracterizados como imuno-estimuladores, são as β-glucanas,
que foram avaliadas em aproximadamente $480 milhões no mercado global em 2020. Suas
aplicações no mercado abrangem seu potencial imunomodulador e sua característica de
alimento funcional. Polissacarídeos e oligossacarídeos adquiriram um espaço dentro do
conceito de prebiótico, por serem fibras não digestíveis que podem ser benéficas ao hospedeiro
ao, seletivamente, estimular o crescimento e/ou atividade de populações da microbiota no
cólon. Essa modulação da microbiota está associada a alteração na função da resposta imune e
a melhora da integridade do microbioma intestinal.
Quando esse microbioma é perturbado, pode ocasionar uma disbiose intestinal que pode
progredir para uma resposta imune crônica tecidual. Localmente, a mudança de população leva
a produção de metabólitos que interfere na homeostase tecidual para um quadro de dano ao
hospedeiro, chamado de colite ulcerosa. Dados anteriores do grupo de pesquisa indicam que
polissacarídeos obtidos do cultivo submerso deste cogumelo atuam na indução de uma resposta
imune protetora na infecção pulmonar pelo fungo Cryptococcus neoformans. Nesse contexto,
um dos objetivos deste trabalho foi avaliar como a administração de exopolissacarídeos obtidos
do macrofungo Auricularia auricula por gavagem em camundongos c57BL/6J, de linhagem
selvagem e Knock-out para o receptor Dectina-1, atuaria em uma modelo experimental de
colite, induzida por dextran sulfato de sódio (DSS). Os resultados obtidos indicam que o
tratamento amenizou os sintomas clínicos da colite, como o encurtamento do tamanho do
intestino e parâmetros histopatológicos e que a resposta é dependente do receptor Dectina-1,
uma vez que os mesmos dados não foram observados nos animais Knock-out para este receptor.
Devido ao baixo rendimento, foi proposto uma metodologia alternativa de extração para
aumentar o rendimento de polissacarídeos a partir da sua purificação do micélio, e observamos
um aumento do rendimento dos polissacarídeos e a sua caracterização mostrou a presença
majoritária de galactose, glicose, manose, com menores quantidades de fucose, xilose e ribose.
Quando comparamos os polissacarídeos obtidos nos dois métodos de extração, observamos que,
na composição monossacarídica predominante, houve um aumento na concentração de
galactose, manutenção da quantidade de glicose e menor quantidade de manose. Analisando a
atividade biológica in vitro deste polissacarídeo observamos que ele foi capaz de estimular a
produção de TNF-α pelos macrófagos. Considerando a capacidade de modular o microbioma
intestinal, analisamos também a capacidade destes polissacarídeos de modular uma infecção
em um outro tecido, e usamos o modelo in vivo do modelo experimental de infecção por
Cryptococcus neoformans. Os animais infectados apresentaram uma disseminação maior do
fungo quando comparado com os animais não tratados ou os Knock-outs para Dectina-1.
Concluindo que apesar de serem extraídos do mesmo macrofungo e ativaram produção
de TNF-α in vitro, os polissacarídeos obtidos do meio de cultura conseguiram controlar um
processo inflamatório in situ enquanto aqueles extraídos do micélio não controlaram a
disseminação fúngica para o sistema nervoso central. | pt_BR |

| dc.description.abstract1 | The use of mushrooms as beneficial agents for human health dates back thousands of
years, especially in Asian culture. The prospect of using polysaccharides, investigated as
potential mediators of different immune responses associated with inflammation and infection
systems, has been the focus of research in recent years. Recent studies demonstrate that the
basidiomycete Auricularia auricula, considered the third most important edible mushroom
cultivated in the world, is used not only in the food context, such as functional foods, but also
presents value for the pharmaceutical industry for its bioactive compounds, such as
polysaccharides, vitamins, proteins, polyphenols, and minerals. Polysaccharides obtained from
this mushroom have antioxidant, antitumor, anticoagulant, hypoglycemic, radioprotective,
antiviral and immunomodulatory activity.
The modulation of the immune response by polysaccharides is well described in the
literature, with the action of cell receptors responsible for binding sugars that lead to cell
activation and signaling, triggering an innate and adaptive immune response. The characteristic
of the response, however, is associated with the molecular and structural composition of these
molecules, which may vary with the monosaccharide composition, branching pattern,
molecular weight, solubility, extraction method of the molecule and the degree of purity
obtained.
One kind of polysaccharides characterized as immunostimulants are β-glucans, which
were valued at approximately $480 million in the global market in 2020. Their market
applications include their immunomodulatory potential and their characteristic of functional
food. Polysaccharides and oligosaccharides have acquired a space within the prebiotic concept,
as they are non-digestible fibersthat can be beneficial to the host by selectively stimulating the
growth and/or activity of microbial populations in the colon. This modulation of the microbiota
is associated with a change in the function of the immune response and an improvement in the
integrity of the intestinal microbiome.
When this microbiome is disturbed, it can lead to gut dysbiosis that can progress to a
chronic tissue immune response. Locally, the change in population leads to the production of
metabolites that interfere with tissue homeostasis leading to a condition of damage to the host,
called ulcerative colitis. Previous data from the research group indicate that polysaccharides
obtained from submerged cultivation of this mushroom act in the induction of a protective
immune response in pulmonary infection by the fungus Cryptococcus neoformans. In this
context, one of the objectives of this work was to evaluate how the administration of
exopolysaccharides obtained from the macrofungus Auricularia auricula by gavage in
c57BL/6J mice, wild type, and Knock-out for the Dectin-1 receptor, would act in an
experimental model of colitis, induced by sodium dextran sulfate (DSS). The results obtained
indicate that the treatment alleviated the clinical symptoms of colitis, such as the shortening of
the intestine size and histopathological parameters and that the response is dependent on the
Dectin-1 receptor, since the same data were not observed in the Knock-out animals for this
receiver.
Due to the low yield, an alternative extraction methodology was proposed to increase
the yield of polysaccharides from their purification from the mycelium, and we observed an
increase in the yield of polysaccharides and their characterization showed the majority presence
of galactose, glucose, mannose, with smaller amounts of fucose, xylose and ribose. When
comparing the polysaccharides obtained in the two extraction methods, we observed that, in the
predominant monosaccharide composition, there was an increase in the concentration of
galactose, maintenance of the amount of glucose and less amount of mannose. Analysing the
in vitro biological activity of this polysaccharide, we observed that it was able to stimulate the
production of TNF-α by macrophages. Considering the ability to modulate the intestinal
microbiome, we also analysed the ability of these polysaccharides to modulate an infection in
another tissue, and we used the in vivo model of the experimental model of Cryptococcus
neoformans infection. Infected animals showed greater spread of the fungus when compared to
untreated animals or Dectin-1 Knock-out.
Altogether, these results show that despite being extracted from the same macrofungus
and activating TNF-α production in vitro, the polysaccharides obtained from the culture
medium were able to control an inflammatory process in situ while those extracted from the
mycelium did not control fungal dissemination to the central nervous system. | pt_BR |
