| Abstract: | The search for treatment of ATL with drugs of easy administration, low toxicity and with greater therapeutic efficacy is the object of studies aimed at curing the disease. The lack of response in some cases and the adverse effects generated the need to use second-line drugs, drug repositioning, topical therapies, and drug combinations in order to increase the effectiveness of the treatment. Given this scenario, the objective of this study was to evaluate the efficacy, adverse effects and immunomodulatory action of the association of oral miltefosine (M) with pentoxifylline (P) and photodynamic therapy with Magonia pubescens hydrogel loaded with C-dots (PDT) in the treatment of CL in BALB/c mice infected with L. (V.) braziliensis. For this, the methodology was based on an in vivo experimental study, in which 54 female mice of the Mus musculus species were divided into nine groups and treated with the following therapies and associations: M, P, PDT, MP, PTFD, MTFD and MPTFD. Efficacy evaluation criteria were aspirated from lesions, investigation of amastigotes, serial dilution and MTT, quantification of parasites by qPCR, measurement of paws, animal weighing, biochemical and immunological tests. The results showed that the MPTFD association was relatively satisfactory after six days of treatment. PTFD combination appears to be more effective than P-only treatment, and also appears to be more effective than PDT-only treatment. The MP combination appears to be more effective than miltefosine alone. Pentoxifylline alone showed similar results to the CP group. PDT appears to be more effective than pentoxifylline treatment. qPCR proved to be efficient to quantify the number of parasites. The association P+PDT, regarding the production of nitric oxide, showed significant relevance and immunomodulatory influence, in the treatment of BALB/c mice infected with L. braziliensis. In conclusion, the importance of the association of drugs and new methodologies is emphasized, given the possibility of increasing the chances of cure in ATL. However, it is recommended in future works the comparison of the MTT technique in relation to qPCR, in order to reassess whether the reduced number of animals may have influenced different results, especially with regard to the sensitivity of the method. |
