| Abstract: | Gliomas are tumors originated from glial cells and included in a heterogeneous group of tumors, with glioblastoma multiforme (GBM) being the most aggressive and having the worst prognosis. GBM grade IV is characterized by being polyclonal and of great clinical and morphological heterogeneity, with great tumor recurrence and resistance to treatments. Even today, understanding the genesis of tumors in the Nervous System is a challenge that makes it more difficult to obtain safer therapies and more effective treatments, therefore, numerous researches within this theme seek to characterize the tumor stem cell in order to clarify questions about the biology of the tumor and its treatment. The glycoprotein CD90 (Thy-1) is a cellular marker present in stem cells of glioblastomas and also in other types of cancer and non-tumorigenic cells. It is known that this molecule is involved with adhesion, proliferation and migration processes and is related to the oncogenesis of some tumors, being pointed out as a promising stem cell marker for these tumors, however, little is known about the relationship between CD90 and the function morphology of glioblastomas. Thus, this research aimed to establish the relationship between the CD90 marker and the tumorigenic and differentiating morphology and capacity invitro. Objective: To obtain primary human glioblastoma cells with reduced CD90 expression to investigate the role of CD90 in the differentiation potential of glioblastoma cells. Methodology: The function of CD90 was evaluated through the morphological and potential characterization of osteogenic, adipogenic and neuronal differentiations, using specific inducing means, in primary cells of glioblastoma multiforme. A population of cells was collected for study by reducing the expression of CD90, using lentiviral vectors that express shRNACD90, and another subpopulation of this, through the separation of cells using a magnetic and anti-CD90 column, in addition to control groups for each analysis. Results: Our work showed that the progressive reduction of CD90 expression resulted in less efficiency in the capacities of neurosphere formation and osteogenic and adipogenic differentiation of these cells. Surprisingly, we found that CD90 ablation favors neuron differentiation from glioblastoma cells. Conclusion: The study suggests that CD90 negatively affects the ability of glioblastoma stem cells to generate a tumor and also affects their ability to differentiate. |
