| Abstract: | Endophytic fungi are microorganisms able to colonize the interior of plant hosts in intercellular spaces or within cells, maintaining a symbiotic relationship. Endophytic fungi can be isolated from several plant species. Their biotechnological potential as enzyme producers has gained more interest in the last decade. In this work, two endophytic fungi were isolated from Solanum falciforme Farruggia, native to Brazilian Cerrado. Isolates, “P1” and “P2” were subjected to molecular identification, using the ITS region to establish their phylogenetic relationship, isolate “P1” was clustered with Diaporthe stictica, while “P2” was clustered with Diaporthe maritima. The isolates were identified as belonging to the genus Diaporthe, reported as one of the most frequently isolated genera of endophytic fungi. They were analyzed for their ability to produce oxidative enzymes: laccases and total peroxidases. Curves of 30 and 15 days were performed to establish the best day of production of these enzymes for each isolate. For “P1”, the 11th day was selected (38.63 Ulaccase/mL; 46.77 Utotal peroxidases/mL) and for “P2” the 10th day (P2: 41.21 Ulaccase/mL; 13.55 Utotal peroxidases/mL). For each isolate a 23 experimental design was carried out with a central point in triplicate to optimize enzymatic production, testing the following factors: initial pH of the medium at levels 4 and 8, amount of peptone, as nitrogen source, at levels 0g and 1g and sugarcane bagasse, as carbon source, at levels 0.25g and 0.75g; central point combined initial pH, peptone and sugarcane bagasse at levels 6, 1.0g and 0.50g, respectively. No model established by the experimental designs obtained statistical significance according to the levels at which these factors were tested (confidence level: 95%), only the factors initial pH, peptone and the interaction between them were considered significant (p <0.05) for the production of laccase for the “P1” isolate. In general, enzyme production increased when the initial pH was at the lowest level and the amount of peptone at the highest level. This work generated relevant data on the extracellular production of oxidative enzymes by endophytic fungi of the genus Diaporthe. |
