Abstract: Cotton is largely cultivated in many countries and stands out for being a source of natural and high-quality fiber for textile industry. The adoption of transgenic variety led to many benefits for farmers, however the cotton genetic transformation process is still a limiting factor. In this way, isolation and characterization of new promoters constitute an essential step in the process of obtaining pest-resistant plants. In this study, we first characterized the constitutive promoter act2 in cotton flowers and bolls and then, ten promoters from tissue-specific genes identified in cotton and Arabidopsis thaliana. For the act2 gene promoter characterization, we used plants (lineage 20/4) from stable transformation previously performed. GUS histochemical staining and fluorometric assays were performed with cotton flowers and bolls. The isolation and characterization of ten tissue- specific promoters were realized in two steps. The first, realized in Mato Grosso Institute of Cotton, included the cotton tissues collection and RNA sample prepare. In Evogene, a gene dataset was selected and characterized in silico using the RePack platform. The genes with preferential expression in reproductive tissues and low or no expression in vegetative tissues were choose and, from them, ten promoters were isolated. The sequences were cloned into plant transformation vectors controlling gus expression. The histochemical assays were carried out using A. thaliana and cotton plants. The act2 promoter was capable to drive high level of gene expression in cotton flower and boll tissues. For the ten tissue-specific promoters, no relation was observed between the expression profile in A. thaliana and cotton tissues. The IMA 001 promoter presented constitutive profile in cotton, similar to CaMV35S. The IMA 006 and IMA 010 had the highest activity in reproductive tissues while showed low or no expression in vegetative and root tissues, which suggests that they may be suitable alternative for use in genetic constructs that target cotton boll weevil resistance. However, considering all the alternatives evaluated in this study, the act 2 promoter was the one that drove the highest expression in the desired tissues.
