| Abstract: | Computer simulation and mathematical modeling of drug delivery systems are fields with enormous potential in scientific innovation and for researching and development of new drugs. The development of drugs demands intense experimental efforts on the phenomena of release, diffusion, absorption and pharmacokinetics, whose costs and execution time can be substantially reduced by using computational modeling. In addition, the safety and efficacy, particularly of potent medicines with narrow therapeutic windows, can be better assured using Controlled Drug Delivery Systems In this Thesis we review the literature on computational modeling of controlled drug release, including works from Takeru Higuchi, Nicholas A. Peppas, Kosmas Kosmidis, J. Siepmann, Peppas, Rafael Villalobos, Gomes Filho, among others. We performed computacional investigations on these system using Monte Carlo simulations of lattice models that were designed to describe drug release from capsule devices with porous membrane. Our study continues the previous investigations by Gomes Filho, where the relationship between Weibull function parameters and some physical characteristics of the system (size, porosity, dimensionality) were observed in one-dimensional, square and cubic porous capsules. We continued the study, however, computationally modeling the behavior of controlled release for encapsulated devices with porous membranes and in different geometries, evaluating the effect of distortion in capsules in two and in three dimensions, in order to verify the effects of distortion on the parameters release. We found that for simulated devices there is a characteristic release behavior, with a similar curve shape resulting from the simulations for different capsule sizes, regardless of dimensionality. Porosity changes also does not change the shape of the release curves. We also evaluated the effects of the variation of porosity on the τ and b parameters of the Weibull adjustment in distorted two-dimensional and three-dimensional capsules, as well as the behavior of the τ and b parameters of the Weibull adjustment, where in the capsule distortion scans of simulated devices in which it remains that the τ parameter presents a behavior of possible convergence by increasing the distortion, while the parameter b varies little, with fluctuation basically centesimal. It is important to highlight that this work consolidates a detailed modeling processes, and can be used as a an initial theoretical framework that may allow, in the future, predictions on feasibility of using certain porous encapsulated pharmaceutical devices to meet the therapeutic windows necessary for their medicinal use. |
