| Abstract: | Alopecia is a disease that affects the hair follicles, gradually decreasing the production of hair shafts. Latanoprost has recently been shown to be a promising molecule for the treatment of this disease, given its influence on hair follicle cells. Considering that the high lipophilicity of this molecule hinders its incorporation in an aqueous-based formulation and restricts its cutaneous penetration, this work aimed to develop a formulation based on polymeric nanocapsules of polycaprolactone incorporating latanoprost in an attempt to promote the accumulation of this substance in hair follicles to favor topical alopecia therapy. The nanocapsules were obtained following the solvent evaporation method. The produced nanosystems had their average size, polydispersity index (PdI) and zeta potential measured through the analysis of light scattering and electrophoretic mobility. The encapsulation efficiency values were determined by quantifying the non-encapsulated latanoprost separated by centrifugation. The stability of the developed nanosystem was monitored for 90 days, observing characteristics such as average size, polydispersity index, zeta potential and drug content. For permeation studies, diffusion cells were assembled with pig ear skin separating the donor compartment from the recipient behavior. Nanocapsules and control formulations were applied for 8 h on the skin surface. The experiments were carried out with and without mechanical stimulation on the skin after the formulation was applied. Nanocapsules of 197.0 ± 2.2 nm (PdI = 0.15 ± 0.01), with zeta potential of -30.1 ± 1.8 mV and capable of encapsulating 93.9 ± 0.4% of latanoprost were obtained. They were stable for at least 90 days when stored at 6oC (209.8 ± 2.31 nm; PdI = 0.35 ± 0.04; zeta potential of -29.2 ± 0.25). Permeation studies showed there was an increase of approximately 20% in the accumulation of latanoprost in hair follicles in the application of nanocapsules’ formulation compared to the control formulation. This difference increased to 86% when the nanocapsules were applied to the skin under a manual massage. Thus, considering that the colloidal dispersion of nanocapsules also demonstrated a very low irritating potential for the skin, it can be concluded that the developed system has a high potential for use as a topical formulation for the treatment of androgenic alopecia. |
