| Abstract: | Introduction: Bisphosphonates (BFs) are anti-resorptive drugs widely used in the prevention and treatment of bone diseases. Despite the benefits of BFs, several side effects have been reported. In addition to Medication-Related Osteonecrosis of the Jaw (MRONJ), the most widely discussed adverse event today, BFs are capable of causing dental changes such as delay or inhibition of tooth eruption and malformations in these structures. Thus, research on the effects of BFs on root structures is important. Objectives: The aim of the present study is to observe, descriptively, the root morphological aspects of molar teeth of Wistar rats submitted to treatment with injectable bisphosphonate (zoledronic acid) and treated with ozone therapy through scanning electron microscopy images. Methodology: At D0, 44 male Wistar rats, approximately 60 days old, were randomly assigned to two groups (n = 22): bisphosphonate treatment (group Z) and control (group C). Each group was subdivided into two subgroups (n = 11): treatment with ozone therapy (OZ group) and no treatment (ST group). In D12, animals from the ZOZ and COZ groups started receiving ozone therapy (20μg/ml) via rectal insufflation at a dose of 5ml, twice a week for 9 weeks. In D21, bisphosphonate therapy (4mg/100ml) of the ZOZ and ZST groups was initiated via intraperitoneal injection at a dose of 66μg/kg once a week for 8 weeks. The COZ and CST groups followed the same protocol, however, with saline injection. At D42, coinciding with the fourth week of drug therapy, the left lower first molars were extracted from all animals. The processing of the samples consisted of metallization of the dental surface with larger area of root exposure for analysis by scanning electron microscope (SEM). For all samples, images were generated with increases of 15, 85 and 200 times. Results: Shallow resorption on the cementum surface was identified in the groups ZST, COZ and CST, predominantly in the root cervical third; cracked regions, at different lengths and depths, on the root surfaces of all groups, predominantly in furcation and root body areas; and cementum of the portions closest to the root apex of all groups with a rougher and voluminous structure when compared to the root body cementum. Moreover, in all groups, root fractures occurred with total involvement of one or more roots. Conclusion: There is insufficient evidence that BFs periodic administration may cause morphological changes in the root topography of rat molar teeth, at least superficially. Additionally, ozone therapy did not prove beneficial or harmful to root surface integrity. Thus, further research is needed to definitively establish the safe use of BFs for dental structures. |
